论文摘要: Cyanophages are widely distributed viruses that specifically infect blue-green algae and play a critical role biological control agents in aquatic ecosystems. Despite their ecological importance, the effects of light on cyanophage-host interactions are not fully understood. This study aimed to investigate the role of host photosynthesis in different stages of MaMV-DH01 infection, a novel muscle-tailed cyanophage isolated from Donghu Lake that targets Microcystis aeruginosa FACHB524. Transcriptomic analysis revealed the significant involvement of host photosynthesis-related genes in the viral life cycle. Differentially expressed genes (DEGs) were validated by quantitative reverse transcription PCR (qRT-PCR), and the impact of photosynthesis inhibitors (N-(3,4-dichlorophenyl)-N-dimethylurea (DCMU) and carbonyl cyanide m-chlorophenyl hydrazone (CCCP)) on viral infection was assessed. The results showed that MaMV-DH01 replication was significantly limited under dark conditions, although light did not affect its adsorption to the host. A large number of DEGs were identified, with the photosynthesis pathway showing the highest enrichment. The use of photosynthesis inhibitors confirmed that disrupting host energy production severely inhibited viral replication. These findings underscored the crucial role of host photosynthetic activity in supporting MaMV-DH01 replication and highlighted the interplay between viral life cycles and host metabolism. This study provided valuable insights into cyanophage-host dynamics and offered a theoretical basis for using MaMV-DH01 in controlling Microcystis aeruginosa blooms under light conditions.
